The .600 nitro express is used for big game hunting in Africa like elephants and water buffalo...etc.
Your .45 isn't gonna do anything more than to piss off a big animal.
The .50 cal is used in the military & .50 BMG round is used by snipers as well.
What kind of idiot would hunt big game with a handgun !?!? that's rifle category, I really don't think you'd want to get close enough to a water buffalo for a handgun to be effective, when you could easily pick him off from a few hundred yards.

bottom line is most people can't handle the kickback/recoil from anything over a .45, I got pushed about a foot and a half back when I was shooting the Desert Eagle .50 cal in vegas, and at the time I was a juiced up 260lbs.
