Yeah, we all know that the compound multiple-joint exercises are real killers ie. Bench Press, Row, Squat, but which single-joint isolation exercise is the hardest? I gotta go wit Stiff-legged Deadlifts.

stiff legged deadlifts are not an isolation movement, they hit spinal erectors, hams, glutes etc. But I see you put "isolation" in quotes so I'm taking it you are casting a wide net.
Anyhoo, if by hardest you mean most painfull, I might suggest seated calf raises. A set of very high rep calf exrecise can really hurt. :'(

That's correct, they ain't really an isolation movement in the sense that they involve multiple muscle groups, but they are a single-joint movement, and some say that single-joint movements are "isolatiom movements". Ok, so how about the hardest and most effective single-joint movement, which one is? I'd say SLD's, but as a very pure isolation single-joint movement is concerned, I would say shrugs.

DJ, if by "isolation movement" you mean revolves around only one axis of rotation, then I guess you could call a SLDL an "isolation movement", but I would never characterize that as such, because the grip is involved, when one is trying to work their hamstrings and low back.

Not if you use straps SilverSpoon ;) And yes, Leg Extensions are rough, but I wouldn't know that much about it, seeing as I hardly ever train my legs ;D

I would have to go with any form of calf exercise - standing, seated, or toe presses on the leg press machine - they all isolate the calves alone and the pain within them when trained to failure can't be compared to any other muscle group.

Because of the length and weight of the legs, you're working at a tremendous leverage disadvantage. Imagine trying to do them strictly with 20lb weights attached to each ankle.

Especially if you do forced reps, breakdowns, and negatives with the leg extensions ;D

If you used the same weight for hanging leg raises as you did for leg extensions, you wouldn't be able to budge the weight.
