Researchers Hail Stem Cells Safe for Human Use

Stem-cell science is a fast-moving field. Just three years since a Japanese researcher first reprogrammed ordinary skin cells into stem cells without the use of embryos, scientists at a Massachusetts biotech company have repeated the feat, only this time with a new method that creates the first stem cells safe enough for human use. The achievement brings the potentially lifesaving technology one step closer to real treatments for disease.

Dr. Robert Lanza, chief scientific officer at Advanced Cell Technology (ACT), reported today in the journal Cell that his team has created stem cells using human skin cells and four proteins. The innovation builds on the breakthrough discovery in 2006 by Shinya Yamanaka, who similarly coaxed human skin cells to revert to a pristine, embryonic state by introducing four key genes into the cells, piggybacked on viruses. However, some of those genes are known to cause cancer, which made Yamanaka's stem cells — known as induced pluripotent stem cells, or iPS cells — unsuitable for human use.

Until now, no one has been able to create iPS cells without using viruses and genes. But Lanza and his team found a way around both: the researchers isolated the proteins made by the same genes Yamanaka used and "tagged" them with a message that allowed the proteins to slip easily into the cell. Yamanaka's method, on the other hand, relied on using viruses to ferry genes into the skin cell.

So far, the ACT group says the iPS cells generated using their method are equivalent to those made using Yamanaka's strategy. In fact, they seem to flourish more robustly than the traditional iPS cells, at least in lab tests that involve regrowing certain cells found in the retina. This could lead to a potential treatment to replace damaged eye cells in conditions such as retinitis pigmentosa or macular degeneration.

Dr. Arnold Kriegstein, director of the Institute for Regeneration Medicine at the University of California, San Francisco, is excited by the advance but notes that it is still relatively inefficient. "The ability to make cell lines from patients with any risk of genetic alteration is a significant step, but the efficiency of 0.001% of cells is low, and even that took two months to achieve," he says, referring to the fact that only a tiny percentage of skin cells in the study transformed into iPS cells over two months. "How readily or quickly this technology is applied, and whether the efficiency is improved, are things that we will have to wait and see."

Still, the rapid pace of the research means we may not have to wait long. "We really lucked out," says Lanza. "These iPS cells were just discovered a few years ago, and here we are three years later with a method safe enough to actually use in people." ACT plans to file a request for the first human trial using its cells sometime next year.

The findings may herald a practical solution to the problem of embryonic stem cells, the controversial cells that can be derived only from embryos (researchers currently use embryos that have been discarded during fertility treatments). President Obama recently lifted the long-standing ban on federal funding of human-embryonic-stem-cell research, but still, these stem cells are unlikely to prove useful as human treatments — for maladies like diabetes, Parkinson's or spinal-cord injuries — since they would not be tissue-matched to the individual patients who need them. The new method could allow scientists to create stem cells using a patient's own skin cells, eliminating the possibility of rejection.

While the new iPS cells may be safer, even Lanza admits there is much that researchers still need to learn about how they will react once inside a patient's body. The most pressing question remains whether iPS cells — or the nerves, muscles or any of the 200 other types of tissues potentially generated from them — would act the same way as human embryonic stem cells, which were not created in a petri dish. "We don't know if iPS cells can do everything that normal human embryonic stem cells can do," says Lanza. "There are certainly a lot of questions we still have to answer." But having a population of stem cells that are safe for human use is an important step toward finding some of those answers.

I don't understand this, becasue the Germans created STEM CELLS from fat (stem cells can be made from Fat, Marrow, Blood, Skin, and quite a few other tissue in the human body) and used them to create stem cells to grow new bone on the skull of a girl who hand a enormous amount of bone loss after an accident and subsequent surgeries could not correct the problem http://www.msnbc.msn.com/id/6727466/

Can you say more american corruption and greedy doctors looking to make money off of these therapies.

but yes, stem cell research around the world is far ahead of the USA. Many US stem cell researchers have been actually doing stem cell therapies outside of the uS in countries where the work is not banned. You seem to be confusing the matter, Jaguar.

The article you linked talks about scientists converting ADULT stem cells, to the type of cells, that supposedly only came from embryonic stem cells, making the use of embryonic stem cells UNNECESSARY.

Stem cell research wasn't BANNED in this country. The only thing Bush did was ban federal funding to embryonic stem cell research. That's the ban that Obama lifted. Embryonic stem cell research (and the other forms) were still legal here.

The reason why embryonic stem cell supporters begged Obama for the money is because US biotech companies won't touch it with a ten-foot pole. Why?

As the saying goes, "If it don't make dollars, it don't make sense". Adult stem cell research has cured/treated over 70 different diseases and ailments; embryonic stem cell research has cured ZILCH!

Conservatives fully support ALL forms of stem cell reserarch, EXCEPT the embryonic kind. Bush's actions send a message: Find a way to get the job done, WITHOUT destroying embryos. It appears that the Japanese have beat us to the punch.

The point: Embryonic stem cell research has too much ethical controversy, costs too much money, hasn't cured JACK, and now has been shown NOT to be the lone source of those precious pluripotent stem cells.

Why waste money on it?

I am a huge supporter of all stem cell research.  In fact, I had stem cells injected into my patellar tendon to attempt to make a chronic tendon injury better.  It absolutely worked.  They took stem cells from my own bone marrow, grew them in culture, and injected them into my tendon.  Over a period of 3-4 months, I had definite improvement in the sx such that I rarely have knee pain anymore.  I would say it was 90% effective.  I could actually go back for another treatment which would likely make it even better, but it does not bother me enough to do it.

If this process eliminates the need for embryonic stem cells, thats great.  IMO, whatever works the best.  Not all of us are so certain of the afterlife and want to make our lives here as full, active and happy as possible.  That includes curing others of disease and disabilities.

Embryos created at the request, and financial and emotional costs of couples attempting to conceive should have full rights to decide what to do with unused embryos -- even permit them to be used to stem cell harvesting and research.  The idea of having their "baby" defrosted on a lab surface to die, rather than contribute to the "good of mankind" has been a spoken issue for parents whose embryos fate was decided for them.
 
Without private business and investors willing to take risks, we would have fewer medications, treatments, and technology than we have available to us today.  There would be less competition to find better and more affordable ways to manage disease.  To demonize the privatization of stem cell research is unfounded:  even government and "not-for-profit" science still must generate success in order to receive continued funding and recognition.  The notion that simply because a business seeks to make a profit makes it incapable of ethical practices and abiding by government regulations is like saying just because you work for a paycheck -- for personal profit -- that you will not be a good employee.  Money is not evil, and not all those skilled in business lack morals and ethics.  Look at Bill and Melinda Gates, Lee Iococa, or many of the other successful businesses that give back generously.
 
The stage at which human embryos are harvested (the blastocyst stage) occurs about 5-7 days after conception.  At this point in development, the blastocyst consists only of a mass of cells and a personal identity as not yet been decided.  From a biological point of view, this is still a stage of human cells, not of a human.  Cells are the building blocks that differentiate (specialize) to form organs, tissues and systems.  These things are what make up a human.  Cells that have not even formed into organs are tissues are pre-human, at best.
 
The Catholic Church states that life begins at conception.  They also state that life is chosen by God and therefore prohibit the use of any birth control -- even the rhythm method.  They teach that the use of birth control keeps a person from being born.  Do we then need to address the rights of eggs or sperm being denied a chance at life when a couple decides to use birth control?
 
The Catholic Church also (currently) forbids the use of condoms -- even between married couples when one partner may be HIV infected.  Not all religious decisions intended to "save" lives do just that.  Some "moral" proclamations, like forbidding condoms, actually result in the loss of life.
 
If it is true that life begins at conception, then it is only fair to completely abolish in vitro fertilization because embryos are created knowing that most will never survive and those unused will either be frozen or thrown away.  If using am embryo for research is "killing" then why is it not killing to discard embryos, or implant 2 or 3 at a time in hope that even 1 will survive?
 
Cord blood, and adult stem cells are older cells.  They grow more slowly than do embryonic stem cells, and are harder to isolate.  While embryonic cells have almost unlimited potential to differentiate, adult stem cells do not, and not all adult stem cells have yet been isolated for all tissues of the body.
 
Adult stem cells are often only present in very small quantities.  Embryonic stem cells are more plentiful, easier to identify, isolate, purify, and are not only more plastic (easier to manipulate) that are also easier to grow.

One, I'm not Catholic.

Two, in true knucke-headed fashion, you continue to perpetuate the myth that conservatives don't support stem cell research. Once again, there's only one form of such that they don't support: embryonic

Three, private investors don't foolishly throw away their money on something they know doesn't work (Reason: NO PROFIT!!!). That's why, genius, embryonic stem cell supporters have to bow down and BEG Obama to fund it for them.

Embryonic stem cell research, to date, has cured absolutely, postively JACK SQUAT!!

Deal with that, before you mindlessly cut-and-paste gibberish from some left-winged site, bashing the Catholic church, without even citing the source.
